I like them both but If I can only choose from the two you have listed I would choose Provolone. Now if I can chose outside the box I would go with Pepper Jack.
Of those choices, definitely American. However, the tuna melt is one of the very few sandwiches where Velveeta is truly the cheese of choice.

I like swiss or provolone on tuna. Make sure it is Kraft or better. Many bargain cheeses are actually congealed cooking oil with coloring added. That is what you get most places that serve fast food. They always say "Do you want cheese with that" and I lol and ask if they actually have cheese - they never do.
Provolone, because it is a White cheese, not a cheddar, and white cheeses go better with fish, cheddars go better with beef.
Cheese much like wine (and they go together!) have certain foods they enhance, and ones they over power, or detract from the taste. Provolone is a mild cheese, and it goes well with mild tasting meats. whereas American is bolder, and overpowers delicate tasting meats (fish and PorK),.

A really good sharp cheddar
Tough call --I love both! I prefer the white ameican though. It has a little more creamy texture than the yellow. But provolone defintely would be my second choice!!
