Is it best to let wine glasses air dry?

As long as they are drying with plenty of airflow, they will be fine air drying or with a towel. Just don't let them (or any dishes for that matter) dry upside down on a towel or they will smell musty!

Yes, air dry then wipe with a soft cloth or paper towel if you have spots. But don't tell the health department I told you to do that. They frown on wiping.

no you will get water spots in the glasses then it will be nasty

I used to own a bar & we let all our glasses air dry.

I let all of my wine glasses air dry but upside down on a clean tea towel instead of upright. Keeping them upside down should minimize the water spots you get but if you have a couple, wipe the glass clean with a paper towel before returning them to where you store your wine glasses.
